摘 要:RNA干扰(RNA interference,RNAi)是一种有力的基因沉默工具,在功能基因组学、微生物学、基因表达调控机制研究等领域广泛应用。RNAi可以用于鉴定药物靶标及治疗疾病,尤其是传统治疗手段无效的重大疾病。如何将双链小干扰RNA(siRNA)高效传递至体内靶细胞是RNAi成功用于疾病治疗的关键,也是目前研究的热点。由于病毒载体能高效转导多种细胞成为基因工程和基因治疗的主要载体。目前,越来越多的研究将病毒载体应用于介导RNAi,然而病毒载体存在靶向性差、生物安全性等问题。本文针对这些病毒载体的优劣进行归纳并指出今后改进的方向。RNA inference (RNAi) as a powerful tool for silencing gene, has been widely used in various research fields, such as functiorial genomics, microbiology and regulatory mechanisms of gene expression. Most importantly, RNAi showed a great potential in screening drug targets and treating human diseases, especially for the incurable diseases by traditional methods. To achieve effective treatment of diseases by RNAi-based strategy, the biggest challenge is how to construct suitable delivery systems for transmitting double-stranded small interference have been developed as the main delivery systems in the fields of genetic engineering and gene therapy. Viral vectors were also applied to transmit RNAi in increasing researches. This review will describe the characteristics of several existing viral vectors applied to deliver small interfering RNA and their improved respects.
