摘　　要：目的:探讨反相高效液相色谱-荧光(RP-HPLC-FLD)法用于NM394血药浓度分析时,如何根据仪器响应值的变化,通过适当的线性回归方式,提高测定的准确度。方法:配制NM394标准曲线系列样品,经处理后进样分析,根据标准曲线上各点准确度的变化规律,选择适当的点,拆分数据,用双标准曲线替代加权回归标准曲线,比较两种处理方法结果的准确度。结果:根据标准曲线各点准确度的变异规律选择拆分点,用双标准曲线替代加权回归标准曲线,结果测定数据在0.02～0.08、0.125～2.5μg/ml范围内线性关系良好,标准曲线各点测得的准确度明显优于单条标准曲线和加权回归标准曲线计算结果。结论:在NM394药物浓度分析过程中,用适当方法拆分数据并进行线性回归,测定的准确度优于单条标准曲线加权回归的结果;经推广应用,该方法同样适用于RP-HPLC-FLD法对其他药物的浓度分析。OBJECTIVE ： To improve the accuracy of determination by the response characteristics of the instrument and proper linear regression equation when blood concentration of NM394 were determined by RP-HPLC-FLD. METHODS： A series of blood samples of NM394 were prepared. After sampling, a point was selected and the data of standard curve was grouped, according to the accuracy changes of standard curves. By the point, weighted regression standard curve was replaced by double-standard curve, while the accuracy of two methods was compared. RESULTS ： The point of splitting was selected according to the variation regulari- ty of the accuracy of standard curves, and weighted regression standard curve was replaced by double-standard curve. By compari- son, the accuracy of the points determined by double-standard curve was better than by single or weighted standard curve. CON- CLUSIONS： In the analysis process of NM394 concentration, the standard curve data is grouped and linear regression is carried out by proper method in this paper. The linear ranges of NM394 are 0.02-0.08 μg/ml and 0.125-2.5 μg/ml. The accuracy of dou- ble-standard curve is better than that of single or weighted regression standard curve. This method is also applicable for other drugs during plasma concentration analysis by RP-HPLC-FLD.
