活血疏肝化痰方影响乳腺癌细胞MCF-7增殖活力的体外研究

摘　　要：目的:探讨活血疏肝化痰方原方及其拆方对乳腺癌细胞MCF-7增殖的影响。方法:采用MTT法观察活血疏肝化痰方原方及其拆方的大鼠含药血清对体外培养的人乳腺癌细胞株MCF-7增殖的影响。结果:原方组、活血拆方组、疏肝拆方组及化痰拆方组含药血清作用12 h后,乳腺癌细胞增殖活性逐渐降低,原方组在血清作用24 h细胞增殖活性最低,细胞生长明显受抑制;疏肝组、活血组及化痰组药物增殖低谷先后出现在药物作用48 h后,且各组增殖活性与浓度相关。结论:原方及其拆方组药物血清对乳腺癌细胞MCF-7具有增殖抑制作用,以原方组效应最明显,各组抑制活性均具有时间和浓度依赖性。Objective： To investigate the effect of Shugan Huoxue Huatan Decoction on proliferation on breast cell line MCF-7.Methods： The inhibition effect of medicine serums of Shugan Huoxue Huatan Decoction on proliferation of breast cell line MCF-7 in vitro was observed by MTF assay. Results： The rates of survival cells in Shugan Huoxue Huatan Decoction group, Huoxue party group, Shugan party group and Huatan party group gradually reduced by the action of the medicated serum began for 12 hours. Shugan Huoxue Huatan Decoction owned the lowest rate of survival cells after 24 hours and cell growth obvious inhibition. The lowest rate of survival of Huoxue party group, Shugan party group and Huatan party group be- gan after 48 hours.The proliferation activity related with concentration. Conclusion： The result of MT＇F assay showed that cell proliferation was significantly inhibited by Shugan Huoxue Huatan Decoction and its separate composes medicine serums. Shugan Huoxue Huatan Decoction had the most significantly specific effects. The effects were correlated with serum concentra- tion and treatment time of Shugan Huoxue Huatan Decoction and its separate composes medicine.
