摘  要:目的探讨新生隐球菌荚膜长期保存及染色方法,以方便教学,随时让学生观察到典型新生隐球菌荚膜负染色的形态。方法取新生隐球菌进行小白鼠腹腔接种,吸出腹腔渗出液,用石炭酸固定液杀菌及固定,并用黑色素代替墨汁染色。结果用石炭酸固定液保存的新生隐球菌菌体及荚膜长达20年之久,镜检形态与新鲜标本无异。结论本法可用于教学,黑色素染色法还可用于临床微生物检验。Objective To examine methods of preserving Cryptococcus neoformans for long periods and staining C. neoformans to facilitate teaching and allow students to observe negative staining of a typical Cryptococcus neoformans capsule at a given time. Methods The abdomen of white mice was seeded with C. neoJormans and abdominal exudate was removed. C. neoformans was killed and fixed with a carbolic acid fixative and then stained with melanin in place of ink. Results C. neoformans and the capsule were preserved in carbolic acid fixative for 20 years. There was no differ ence between the observed morphology of preserved specimens and fresh specimens. Conclusion This method of preset vation can be used in teaching, and melanin staining can be used for clinical examination of microorganisms.
