摘 要:为探讨F-box蛋白在小麦发育过程中的调节作用,利用电子克隆技术获得了一个小麦F-box蛋白基因,采用生物信息学方法,对其编码的蛋白质从序列特征、理化性质、跨膜结构域、亚细胞定位、高级结构及功能域等方面进行了预测和分析。结果表明,小麦F-box蛋白由362个氨基酸组成,在N端和C端分别存在一个F-box保守结构域和一个DUF295保守结构域(功能未知),该蛋白质二级结构中以α-螺旋(23.48%)、扩展束(25.41%)和无规则卷曲(47.24%)为主,在94-115位点处可能存在一个由内向外的跨膜区,N端不存在信号肽序列,亚细胞定位于细胞质中;该F-box蛋白与二穗短柄草、玉米和水稻的F-box蛋白相似性较高,进化距离较近。In the paper,an F-box protein gene from wheat was obtained by electronic clone technique,and some characters of the F-box protein were analyzed and predicted by bioinformatics method,including the composition of amino acid sequence, hydrophobicity, secondary structure and function of protein etc. The results showed that the F-box protein was composed of 362 amino acids,and contained an F-box domain in N end and a DUF295 conserved domain in C end of protein sequence. The secondary structure of the F-box protein mainly included a-helix (23.48%), extended strand(25.41% ) and random coil (47.24%). The F-box protein had higher sequece indentities and closer evolution distance with that from other Gramineae plants including Brachypodium distachyon ,Oryza sativa and Zea may.
