摘 要:为了研究光质对铁棍山药试管苗生根的影响,将带1个叶片、长2~3cm的茎段,接种在1/2MS+IBA 0.5mg/L+PP3330.05mg/L培养基上,置于蓝光、绿光、黄光、红光、白光及黑暗下培养。结果表明:接种7d,在黄光下的茎段切口处首先有根点出现,11d各光质下均有根长出。从根数和根长上看,红光表现出促进效应,达到5.02个和6.08cm,而蓝光和绿光则表现出抑制效应。对于根鲜质量、根冠比来说,白光下的比较大,达到1.20g和0.563,而蓝光、绿光下的则比较小。从侧根数来看,白光下侧根最多,而红光下极少。从苗情上看,白光、红光下的试管苗比较矮壮,叶片数较多,叶绿素含量较高。综合看来,白光和红光有利于铁棍山药试管苗的生根。The aim was to study the effects of light qualities on rooting of Tie-gun Dioscorea opposita Thunb. plantlet. In 1/2MS-IBA 0. 5 mg/L-PP333 0.05 mg/L medium, the segments in 2-3 cm length with a leaf were cultivated under blue lights, green lights, yellow lights, red lights, white lights or dark. The results showed that seven days later, roots appeared under the yellow light;eleven days later,roots appeared under every lights. The red light was beneficial to the number and length of roots, with 5.02 roots and 6.08 cm in length, while the blue and green lights gave the inhibitory effect. The white light was beneficial to fresh mass of roots and root top ratio,with 1.20 g in weight and 0. 563,while the blue and green lights were inhibitory for rooting. The white light was beneficial to the branch roots production while the red light had an inhibitory effect. Under the white and red lights, the test-tube plantlets were lower and stronger, with the leaves more and the chlorophyll content higher. In short, the white and red lights were beneficial to roots production of Tie-gun Dioscorea opposita Thunh. test-tube plantlet.
