摘 要:目的观察丙泊酚对心肌肥厚大鼠心肌肌浆网Ca2+-ATPase活性的影响。方法 50只雄性SD大鼠采用SAS统计软件随机分为正常对照组(n=10)和心肌肥厚模型组(n=40)。心肌肥厚组再随机均分四个亚组(n=10):模型组、丙泊酚30mg组、丙泊酚60mg组和脂肪乳剂组。采用腹腔注射去甲肾上腺素(NE)方法制备大鼠心肌肥厚模型,NE 1.5mg.kg-1腹腔注射,每天2次,连续15d得到心肌肥厚模型。次日麻醉后分别静脉输注生理盐水、不同剂量丙泊酚及脂肪乳,输注30mins后处死大鼠,测量心肌组织Ca2+浓度及肌浆网Ca2+-ATPase活性。结果与正常对照组比较肥厚模型组各亚组心肌组织Ca2+浓度增高(P<0.05),而肥厚模型组各亚组比较Ca2+浓度差异无统计学意义(P>0.05)。与正常对照组比较肥厚模型组各亚组心肌肌浆网Ca2+-ATPase活性降低(P<0.05),肥厚模型组各亚组比较:模型组、脂肪乳剂组高于丙泊酚30mg组、丙泊酚60mg组,丙泊酚30mg组高于丙泊酚60mg组(P<0.05),余差异无统计学意义(P>0.05)。结论心肌肥厚大鼠心肌肌浆网Ca2+-ATPase活性与丙泊酚的剂量有关,随着剂量增大活性降低。Objective To investigate the effects of propofol on Ca2+-ATPase activity of myocardial plasma membrane in cardiac hypertrophy rats.Methods Fifty SD rats were randomly divided into normal control group(n=10) and model of myocardial hypertrophy group(n=40).The myocardial hypertrophy group were randomly divided into four subgroups with 10 rats earch: model group,30mg propofol group,60mg propofol group and intralipid group.Forty myocardial hypertrophy model rats were treated with norepinephrine(NE) 1.5mg·kg-1 intraperitoneally twice daily for 15 day.On the 16th day,normal saline,different doses of propofol or intralipid were infused intravenous after anesthesia.Calcium ion concentration and Ca2+-ATPase activity of myocardial plasma membrane were measured after 30 min infusion.Results Compared with normal control group,calcium ion concentration of each subgroups were significantly higher(P0.05),however no statistical significance among subgroups of group B(P0.05).Compared with normal control group,Ca2+-ATPase activity of each subgroups were significantly decreased(P0.05).Among myocardial hypertrophy group : model group and intralipid group were significantly higher than 30mg propofol group and 60mg propofol group which 30mg propofol group was significantly higher than 60mg propofol group(P0.05).Conclusion Cardiac hypertrophy of rat myocardial plasma membrane Ca2+-ATPase activity is related to the dose of propofol,and the activity was reduced as the dose increased.
