摘  要:从新疆硝尔库勒盐湖分离纯化到一株极端嗜盐古菌TRM 51T,其菌落颜色为白色/粉红,细胞为杆状,能运动,革兰氏染色为阴性,生长温度范围是28~50℃(最适温度37℃),NaCl浓度生长范围是2.1~4.3 M(最适浓度为3.1 M),Mg2+生长范围是0.03~1.0 M(最适0.3 M),pH生长范围5.5~9.0(最适pH8.0)。细胞可在蒸馏水中裂解,避免细胞裂解的最低NaCl浓度为8%(w/v)。TRM 51T主要的极性脂是PG(磷脂酰甘油)、PGP-Me(磷脂酰甘油磷酸甲基酯)、TGD-2(葡糖苷甘露糖苷葡糖二醚),基于16S rDNA基因的系统发育分析显示,菌株TRM 51T与相近属Halopiger的成员相似性仅为94.2%~95.3%,与Haloterrigena类群的相似性为92.4%~94.6%,菌株TRM 51T基因组DNA的G+C mol%为63.2mol%。多相分类结果表明,菌株TRM 51T不能归入嗜盐古菌科的已描述属,建议成立新属级的新分类单元。A halophilic archaeal strain TRM511 was isolated from Xiaoerkule salt lake in Xinjiang, China. The colonies were white/ pink and the cells were rod - shaped, motile, and Gram negative. It was able to grow at 28 ~ 50℃ ( optimum 37 ℃, at 2. 1 - 4. 3 M NaC1 (optimum3. 1 MNaC1), at0. 03-1. 0MMgC12(optimum0. 3MMgC12) and atpH5. 5-9. 0 (optimumpH8. 0). Cells lysed in distilled water and the minimal NaC1 concentration to prevent cell - lysis was 8% (w/v). The major polar lipids of strain TRM 51 ^T were PG (phosphatidylglycerol), PGP -Me (phosphatidylglycerol phosphate methyl ester), TGD -2 (glucosyl mannosyl glucosyl diether). Phylogenetic analysis based on 16S rDNA gene revealed that strain TRM51 T was phylogenetically related to Halopiger (with 94. 2% - 95. 3% similarities) and Haloterrigena (92. 4% - 94. 6% ). The DNA G + C content of strain TRM51T is 63. 2 %. It was concluded that strain TRM51T could not he classified in any recognized genus of Archaea Branch. Strain TRM 51T is thus consid- ered to represent a novel species in a new genus within the family Halobacteriaceae.
