摘　　要：目的观察同种异体骨髓间充质干细胞(BMSCs)心肌内移植对60分钟缺血/再灌注心肌梗死(MI)模型大鼠左心室重塑及心肌去甲肾上腺素转运蛋白(NET)表达的影响,探讨BMSCs移植对心脏交感神经功能以及胶原重塑作用的可能机制。方法分离、原代培养Wistar大鼠BMSCs,建立缺血/再灌注MI模型,随机分为假手术对照组、MI组、BMSCs移植组。观察BMSCs移植后动物模型血流动力学指标,天狼猩红染色测定MI范围、面积以及扩展指数,结合偏振光分析MI区胶原容积分数和胶原成熟度;免疫组化检测NET表达。结果①BMSCs移植可缩小60min缺血/再灌注大鼠MI面积;②BMSCs移植可减轻心肌梗死动物左心室重塑;③BMSCs移植发挥细胞外基质抑制,心肌间质胶原含量、胶原容积分数下降,增加心肌NET表达。结论 BMSCs移植可改善MI后心室重塑、降低心肌胶原、增加心肌NET表达。Objective To observe the influences of myocardial transplantation of bone mesenchyme stem cells（BMSCs） on left ventricular remodeling and expression of norepinephrine transporter（NET） in rat model of 60-minute ischemia/reperfusion myocardial infarction（MI）,and investigate the effective mechanism of BMSCs transplantation to cardiac sympathetic function and collagen remodeling.Methods BMSCs were separated for primary culture in Wistar rats,model of ischemia/reperfusion MI was established and randomly divided into sham-operation group,MI group and BMSCs transplantation group.The indexes of hemodynamics were observed after BMSCs transplantation.The range,area and expansion index of MI were determined after Sirius red staining.The collagen volume fraction and collagen maturity in MI area were analyzed combined polarization analysis.The expression of NET was detected by using immunohistochemistry technique.Results ① BMSCs transplantation reduced MI area in 60-minute ischemia/reperfusion rats.② BMSCs transplantation relieved the left ventricular remodeling in MI rats.③ BMSCs transplantation inhibited extracellular matrix,decreased myocardial collagen content and collagen volume fraction,and improved the expression of myocardial NET.Conclusion BMSCs transplantation can relieve post-MI ventricular remodeling,reduce myocardial collage content and improve the expression of myocardial NET.
