摘 要:采用荧光光谱滴定法研究了不同温度下5-对羟基苯基-10,15,20-三甲氧基苯基锌(钴)卟啉与人血清白蛋白(HSA)的结合反应。按照Stern-Volmer方程、Lineweaver-Burk双倒数方程分析和处理实验数据,得到了结合反应的猝灭常数、结合常数、结合位点和热力学参数等。结果表明,上述配合物与HSA之间形成了稳定的1∶1的复合物,并通过氢键或范德华力发生了较强的静态荧光猝灭作用。The interaction between 5-(4-hydroxyl)-10,15,20-triphenylporphyrin zinc (or cobalt) and human serum albumin (HSA) were studied by means of fluorescence spectrum in order to simulate biological function under different temperature. The quenching constant Kq, binding constant KLS, binding site n and thermodynamic parameters of the interaction were obtained based on the analysis of the fluorescence quenching data according to Stem-Volmer equation and double-reciprocal equation. The results showed that the 1 : 1 complexes were formed and there was stronger quenching interaction between metalloporphyrins and HSA by hydrogen bonding or Van der Waals interaction.
