摘 要:目的探讨纳米羟基磷灰石/羧甲基壳聚糖(N-HA/CMCTS)复合材料的细胞相容性,评价其作为软组织填充剂的可行性。方法设计制成复合材料(纳米羟基磷灰石与羧甲基壳聚糖的比例为6∶4),小鼠成纤维细胞L929行体外细胞传代培养。制备材料浸提液,MTT法进行细胞毒性评价,细胞接种于复合材料,通过倒置相差显微镜,应用形态观察法、细胞增值率,定性定量观察复合材料对细胞生长、附着、增值的影响。结果纳米羟基磷灰石/羧甲基壳聚糖(N-HA/CMCTS)复合生物材料的细胞毒性不大于1级。成纤维细胞在复合材料的环境中生长良好并促进细胞生长。结论纳米羟基磷灰石/羧甲基壳聚糖(N-HA/CMCTS)复合材料具有良好的生物相容性,作为一种新型的注射性软组织填充材料有望应用于整形美容领域。Objective To discuss the Cytocompatibility of the N- HA/CMCTS composite, and evaluate the feasibility of it as a filler for soft tissue. Methods The composite was made by pre - designed (N - HA and CMCTS for portion 6 : 4), mouse L929 fibroblasts cells were isolated and multiplied in vitro. The cyto- toxicity of N- HA/CMCTS composite was evaluated through MTT method. The mouse L929 fibroblasts cells were seeded into the composite. The effect of the N- HA/CMCTS composite on the cell adhesion, proliferation and morphological changes were observed by the phase - contrast microscopy. Results The cytotoxicity of the N- HA/CMCTS composite had been graded 0 and the N- HA/CMCTS composite sup- ported L929 cells to cohere and increase well in vitro. Conclusion The N- HA/CMCTS composite has a good biocompatibility and will be as a new injectable soft tissue filler material for the field of cosmetic sur- gery.
