摘  要:目的:观察表达嵌合体蛋白PAcA/CTB转基因番茄防龋疫苗的免疫原性和免疫反应性。方法:新西兰大白兔随机分为实验组、阴性对照组和阳性对照组3组,分别喂食3个剂量(目的蛋白量相差1.5倍)的转基因番茄汁、非转基因番茄汁和变异链球菌灭活全菌疫苗。每周免疫1次,连续免疫4周,ELISA法检测免疫前和免疫后不同时期血液、唾液样品中抗变异链球菌PAc的IgG、IgA抗体效价。结果:口服免疫后实验组与阳性对照组大白兔血液和唾液中均出现特异性的抗PAc抗体,抗体水平可维持数周,而阴性对照组抗体水平变化不明显。结论:表达嵌合体蛋白PAcA/CTB的转基因番茄能够诱导动物机体产生有效的系统免疫应答和黏膜免疫应答。To observe the immunogenic and immunoreactivity of the transgenic tomato anti-caries vaccine which express chimeric protein PAcA/CTB by animal experiment. Methods: New Zealand white rabbits divided into experiment group, positive group and negative group randomly and fed the transgenic tomato juice, non-transgenic tomato juice and inactivated whole cell vaccine of streptococcus mutans with three different dose(the amount of target protein difference 1.5 times). Immune four times , once a week, antiboby titer was analyzed by ELISA to observe the immunogenic antiboby of the IgG and IgA of serum and saliva. Results: The PAC-specific antibodies was appeared in both serum and saliva of experimental group and the positive group's rabbits. The antibody levels could be maintained for several weeks, but the antibody of negative group did not change significantly. Conclusion: The transgenic tomato which express chimeric protein PAcA/CTB effectively induce local mucosal immune response and systemic immune response.
