摘　　要：目的　探讨酪氨酸激酶信息传递途径在成骨细胞增殖及 c- fos表达中的作用。方法　采用阶段性酶消化法分离培养新生大鼠颅盖骨成骨细胞 ,用 MTT法观察成骨细胞增殖 ,用免疫组化 SP法结合计算机图像处理系统检测成骨细胞 c- fos表达。结果　酪氨酸激酶抑制剂 Genistein和 HerbimycinA抑制成骨细胞增殖 ,且两者均部分阻断血清刺激的成骨细胞 c- fos表达。结论　血清刺激和维持成骨细胞增殖及诱导 c-Objective To study the signal transduction pathway in the proliferation and c fos expression of osteoblasts.Methods\ Osteoblastic cells from rat calvaria were isolated by sequential enzymatic digestion.The proliferation of osteoblasts was measured by tetrazolium salt(MTT)method,and c fos expression of osteoblasts was detected by immunohistochemistry combined with computer based image analysis.Results Both genistein and herbimycin A,the inhibitors of tyrosine kinase,inhibited proliferation of osteoblasts,and they both partly blocked the c fos expression induced by fresh serum. Conclusion Tyrosine kinase pathway is involved in the proliferation and c fos expression of osteoblasts induced by serum.
