摘 要:目的观察去细胞组织工程猪肺动脉带瓣管道的血液相容性。方法对新鲜猪肺动脉带瓣管道采用胰蛋白酶+Triton X-100的去细胞方法进行去细胞处理;对去细胞猪肺动脉带瓣管道进行明胶嵌合再处理;对3组(新鲜猪肺动脉组、单纯去细胞处理组及明胶嵌合去细胞再处理组)瓣膜及管壁组织通过动态凝血试验、溶血试验、血小板黏附性能试验,分析3种组织材料的血液相容性。结果相对新鲜猪肺动脉组及单纯去细胞组,明胶嵌合再处理组的瓣膜及管壁组织具有更低的凝血率,同时血小板黏附更少且没有明显变形和伪足。结论采用新型方法构建的组织工程猪肺动脉带瓣管道,具有良好的血液相容性,有望解决移植体内的血栓形成问题,延长其使用寿命。Objective To observe the hemocompatibihty of decellularized porcine tissue-engineering pulmonary valved conduits. Methods Using trypsin + Triton X-100 to handle fresh porcine pulmonary valved conduits, taking off all the cells and extracellular matrix in the valve and wall; re-treating decellularized porcine pulmonary valved conduits by gelatin mosaicing; then they were divided into fresh porcine pulmonary group, simple decellularization group and gelatin mosaic a- cellular porcine pulmonary valved conduits group. The hemocompatibility of these three groups was analyzed by the dynamic blood clotting test, hemolytic test and platelet adhesion performance test. Results Compared with the fresh porcine pulmo- nary group and.decellularization group, valves and wall tissues in gelatin chimeric re-treatment group had a lower coagula- tion rate. At the same time platelet adhesions were much fewer, and had no apparent deformation and pseudopodium. Con- clusions The new gelatin mosaic acellular porcine pulmonary valved conduits have good blood compatibility and they'are expected to solve the problem of graft thrombosis and prolong their service life.
