摘 要:【目的】探讨叶绿体23S-4.5S-5S rDNA内转录间隔区序列在甘蔗近缘属种系统进化中的应用,为深入研究甘蔗近缘属种系统进化提供参考。【方法】以14个甘蔗近缘属种材料为研究对象,测序分析其23S-4.5S-5S rDNA内转录间隔区序列,并应用BLAST分析其在禾本科中的变异情况。【结果】23S-4.5S-5S rDNA内转录间隔区序列在14个甘蔗近缘属种材料间的同源性为100%,属于高度保守区域,但在禾本科各亚科间表现出一定的变异,且在各亚科中都存在其特异的变异位点。【结论】23S-4.5S-5S rDNA内转录间隔区序列在甘蔗近缘属种间属于高度保守的区域,不适合用于甘蔗近缘属种间系统进化的分析,但可为禾本科亚科系统进化的分析提供有用的信息。[Objective ]The application of chloroplast 23S-4.5S-5S rDNA ITS to sugarcane related genera and species was studied to provide references for further research on phyletic evolution of sugarcane related genera and species. [Method]After sequencing the 23S-4.5S-5S rDNA ITS region of 14 accessions of sugarcane related genera and species, its variable sites among accessions in Poaceae were analyzed by BLAST program. [Result]The 23S-4.5S-5S rDNA ITS re- gions, which belonged to highly conservative regions, showed 100% of homology among 14 accessions of sugarcane related genera and species, but still presented variable sites in sub-family level of Poaccae, and all sub-family had its special variable sites. [Conclusion]Because the 23S-4.5S-5S rDNA ITS region was highly conservative, it was unsuitable for phy- logenetic study of sugarcane related genera and species, but it could provide some useful information for phylogenetic study in sub-family level of Poaceae.
