摘 要:就不同消毒方法和不同培养基对日本菟丝子(Cuscuta japonica)茎段消毒与组培效果的影响进行了研究.结果表明,于100 mLφ(乙醇)=75%中浸泡20 s后用无菌水冲洗2次,500 mLω(HgCl2)=1%中加入0.2 mL吐温-80浸泡4 min后再用无菌水冲洗4次,对日本菟丝子茎段消毒的效果最佳;MS+NAA 1.0 mg/L+活性炭(Activated carbon,AC)0.1 mg/L或1/2 MS+NAA 1.0 mg/L+AC 0.1 mg/L是其茎段最佳培养基.利用该培养基组培获得的菟丝子藤蔓出现类似吸器的突起,在培养瓶内能开出正常的花朵,但未观察到有根的生成,也没有出现自然状态下的自身缠绕现象.Sterilization effect and tissue culture of Cuscuta japonica's stem segments were studied by different disinfection methods and media. The results showed that the optimal disinfection method for C. japonica's stem segments were as follows: soaked in 100 mL 75% alcohol, then washed 2 times with sterile water after 20 s, followed by soaking in 500 mL 1% mercuric chloride contained 0. 2 mL Tween-80 for 4 min and washing 4 times with sterile water. The optimal medium was MS + NAA 1.0 mg/L + Activated carbon (AC) 0. 1 mg/L or 1/2 MS + NAA 1.0 mg/L + AC 0. 1 mg/L. The haustoria were observed in the dodder during tissue culture. During the tissue culture, normal flowers were observed in the culture flask, but the root could not be formed, and there was no self-wrapped phenomenon observed at the natural condition.
