硫酸新霉素软膏经大鼠离体烧伤皮肤透皮吸收量的实验研究

摘  要:目的研究硫酸新霉素软膏经完整皮肤,二度烧伤皮肤和三度烧伤皮肤的透皮吸收量的差异,为临床合理使用硫酸新霉素软膏提供参考。方法以完整皮肤为对照组(n=5),二度烧伤与三度烧伤为实验组。采用改良Franz扩散池和大鼠离体皮肤进行体外渗透试验。以茚三酮-吡啶为衍生化试剂,利用反相高效液相色谱法对接收液中的硫酸新霉素进行含量测定,计算硫酸新霉素累积渗透量。结果完整皮肤,二度烧伤及三度烧伤皮肤6h的累积透过量分别是529.872μg,1411.59μg,1901.212μg。二度烧伤皮肤的硫酸新霉素透皮量与完整皮肤透皮量的的差异有统计学意义(P<0.05)。三度烧伤皮肤的硫酸新霉素透皮量与完整皮肤透皮量的透皮量的的差异有统计学意义(P<0.05)。三度烧伤和二度烧伤皮肤的硫酸新霉素透皮量之间的差异也有统计学意义(P<0.05)。结论硫酸新霉素软膏经烧伤皮肤的透皮吸收量比完整皮肤的透皮吸收量较大。OBJECTIVE To research the different transdermal absorption of neomycin sulfate ointment absorbed through the complete, the second degree and the third degree burned skin, provide reference for the rational clinical use of neomycin sulfate ointment. METHODS First, take the complete skin as the control group ( n = 5 ), the second degree and the third degree burned skin as the experimental group;Second, use modified Franz diffusion pool and iso- lated burned-rat skin for penetration test. And third, with acidic ninhydrin-pyridine for derivatization reagent, and with the method of RP-HPLC for the determination of the content of neomycin sulfate in receptor fluid, calculate the cumu- lative permeation rate of neomycin sulfate. RESULTS The cumulative penetration amount of the complete skin, the second degree and the third degree of the 6-hour-urned skin are 529. 872μg, 1411.59μg, 1901. 212μg respective- ly. As a result, the difference of the absorption of neomycin sulfate among these three kinds of skin was statistical sig- nificanee (P 〈0. 05 for all). CONCLUSION The transdermal amount of Neomycin sulfate through the burned skin is larger than through complete skin.
