摘  要:目的:探讨依达拉奉对吸入性肺损伤大鼠的肺保护作用。方法:将40只雄性Sprague-Dawle大鼠采用随机数字表法随机分为4组,每组10只。分别为正常对照组(A组),致伤空白组(B组),致伤依达拉奉8 h治疗组(C组),致伤依达拉奉24 h治疗组(D组)。C组及D组致伤后30 min给予腹腔注射依达拉奉(9 mg/kg),B组致伤后30 min给予等量生理盐水,A组无处理。C组及D组分别于伤后8 h及24 h留取各组大鼠腹主动脉血液样本离心检测肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量,留取肺组织制备肺组织匀浆后测定丙二醛(MDA)含量、超氧化物歧化酶(SOD)、一氧化氮(NO)及诱导型一氧化氮合酶(iNOS)活性。取部分右肺组织做病理HE染色切片光镜观察。结果:与B组相比,C组TNF-α、IL-6、MDA、NO及iNOS水平显著降低(P<0.01),SOD水平增高(P<0.05);D组较B组上述变化更明显(P<0.01)。光镜下C组和D组较B组肺组织水肿减轻,炎性细胞浸润减少。结论:依达拉奉可能通过抑制自由基和部分炎症介质的产生和释放减轻烟雾吸入性肺损伤大鼠体内炎性反应,对肺组织起到一定的保护作用。Objective: To investigate the protective effects of edaravone on lung in smoke inhalation injury in rats. Methods: Thirty-two male SD rats were randomly divided into four groups, Group A: normal control group(n=10), Group B: injured group(n=10), Group C:edar- avone 8 h treated group (n=10), Group D: edaravone 24 h treated group (n=10). Thirty minutes after smoke inhaled injured, the rats in group C and group D were all injected intraperitoneally with edaravone (9 mg/kg), with the same volume the group B were injected in- traperitoneally normal saline. The rats were treated by no means in group A. The blood samples were collected from abdominal aorta and the lung tissue of the rats in group C at 8 hours after injured, and the same to group D at 24 hours after injured. The levels of tumor necrosis factor-ot(TNF-a), interleukin-6(IL-6) in serum as well as the levels of malondialdehyde(MDA), nitric oxide(NO), inducible nitric oxide synthase (iNOS)and superoxide dismutase(SOD) in lung tissue were tested. Each right lung tissues were examined under an optical micro- scope. Results: Compared with group B, the TNF-α, IL-6, MDA, NO, iNOS levels decreased markedly in group C (P〈0.01),while the lev- els of SOD were increased (P〈0.05). The above changes were more evident in Group D (P〈0.01). Histopathological examination showed that inflammatory granulocytes and edema were improved in group C and group D compared with group B. Conelusion:Edaravone has protective effects on smoke inhalation injured, by inhibiting the production and release of free radicals and inflammatory mediators.
