摘 要:目的:探讨槲皮素在吸入性损伤中对肺的保护作用。方法:雄性Sprague-Dawle大鼠40只采用随机数字表法随机分为4组:正常对照组(A组)、致伤空白组(B组)、槲皮素治疗组(C组)、槲皮素预防组(D组),每组10只。C组在致伤后3 h腹腔注射槲皮素(200 mg/kg),D组在致伤前10 min腹腔注射槲皮素(200 mg/kg),B组于致伤后3 h腹腔注射等量的生理盐水,A组无处理。各组分别于致伤后72 h留取大鼠腹主动脉血液样本5 mL离心检测肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)含量,留取肺组织制备肺组织匀浆后测定丙二醛(MDA)含量、超氧化物歧化酶(SOD)及诱导型一氧化氮合酶(iNOS)活性。取右肺组织做肺组织干湿比。取部分肺组织做病理HE染色切片并光镜观察。结果:C组较B组TNF-α、IL-6、MDA、iNOS水平降低(P<0.01),SOD水平增高(P<0.01)。D组上述变化更明显(P<0.01)。光镜下B组肺泡形态不规则,肺泡壁不均匀,大量炎症细胞浸润,C组及D组较B组病理变化减轻。与C组比较,D组肺含水量下降(P<0.05)。结论:槲皮素可能通过增强肺水的清除,减少炎症介质的释放及抑制自由基生成对烟雾吸入性肺损伤起到保护作用。提前用药效果更好。Objective: To explore the protective effects of quercetin on smoke inhalation injury in rats. Methods: Forty male SD rats were randomly divided into four groups. Group A: control group (n=10),Group B: injured group (n=10), Group C quercetin treated group (n=10) and Group D:quercetin prevented group (n=10). Three hours after smoke inhaled injured, the rats in group C were all injected in- traperitoneally with quercetin (200 mg/kg). Ten minutes before smoke inhaled injured, the rats in group D were all injected intraperi- toneally with quercetin(200 mg/kg),at the same time the group B were injected the same volume normal saline. The rats were treated by no measures in control group. At the time of 72 hours after smoke inhaled injured, the blood samples 5 mL were collected from abdominal aorta and the lung tissue. The levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) in serum as well as the levels of malondi- aldehyde (MDA), inducible nitric oxide synthase (iNOS) and superoxide dismutase (SOD) in lung tissue were tested. Lung tissues were checked for moisture and examined under microscope. Results: The levels of TNF-α, IL-6, iNOS and MDA in group C were lower than those in group B (P 〈0.01),and the SOD were higher (P 〈0.01). The above changes were more evident in group D (P〈0.01). Pathology showed thickened alveolar space, inflammatory cells effusion, and local hemorrhage in group B, inflammatory granulocytes were improved in group C and group D. Compared with group C, pulmonary edema was found to be lower in the group D. Conclusion: Quercetin has protective effects on smoke inhalation injury by relieve pulmonary edema, scavenging free radical and inhibiting proinflammatory cy- tokines. The effects of quereetin on smoke inhalation injury is better in prevention.
