摘  要:针对奶牛粪便中的纤维素,进行了高酶活纤维素降解菌的筛选,从奶牛场排水口处土壤、林地和菜园土壤中共分离得到19株纤维素降解菌,经过滤纸条复筛和奶牛粪便纤维素降解率的测定,选出3株降解率相对较高的菌株,分别为F2,F3和F9.对其进行诱变选育,F3.1.6和F9.1.8酶活提高较大,其CMC酶活和滤纸酶活分别提高了47.69%,47.51%和53.41%,50.17%.继代培养后比较可得,F9.1.8传代稳定性较好,其CMC酶活和滤纸酶活较原始菌株F9分别提高了13.47%和22.11%,可作为后续发酵产酶的生产菌株.Aiming at the cellulose in the dairy manure,this paper screens the highly-active cellulose-decomposing microorganisms.In this study,19 cellulose-decomposing microorganisms have been separated from the soil of outlet of a dairy,the forestland on the campus of the Southwest University and the soil of a vegetable garden.Three microorganisms with relatively higher decomposition rates have been selected by the menstruation of cellulose-decomposing rates in dairy manure and the repetition of screening through filter paper and that is F2,F3,and F9.Through mutation breeding and screening,the activity of F3.1.6 and F9.1.8 have increased greatly.Their CMCase and FPase have increased by 47.69%,47.51% and 53.41%,50.17%.After subculture,F9.1.8 shows relatively stable generations and compared to the original strain F9,the CMCase and FPase have increased by 13.47% and 22.11%,which can be the follow-up strain to produce enzyme.
