摘  要:目的探讨甘氨双唑钠对离体乏氧喉癌细胞的放射增敏作用及机制。方法以加药乏氧照射喉癌细胞为实验组,单纯乏氧照射喉癌细胞为对照组;实验组分别给予不同浓度的甘氨双唑钠,经照射后,应用MTT法、克隆形成试验检测其放射增敏作用;并应用流式细胞仪分析放射后细胞周期变化情况。结果 MTT法和克隆形成实验均证明甘氨双唑钠对乏氧喉癌细胞具有显著放射增敏作用,随着药物浓度增加,放射增敏比SER分别为1.23、1.54、1.98,放射增敏作用增强,各组间比较有统计学差异(P<0.05)。流式细胞仪检测结果显示,乏氧喉癌细胞发生G2/M期阻滞,甘氨双唑钠药物浓度越高,G2/M期比例越高,呈上升趋势。结论甘氨双唑钠对离体乏氧喉癌细胞具有明显放射增敏作用,其增敏效果随药物浓度的增加而增强,其放射增敏机制可能与减小对放射耐受的G0/G1期细胞比例、增大对放射敏感的G2/M期细胞比例有关。Objective To investigate the radiosensitizing effect of CMNa on the hypoxic laryngeal carcinoma Hep-2 cell line in vitro. Methods The hypoxic Hep-2 cell lines were divided into radiation group as control group, and CMNa combined with radiantion as experimental group. Then MTT assay and Colon formation method were used to investigate the radiosensitivity of CMNa in laryngeal carcinoma cells. Changes of cell cycle after radiation with different concentration of CMNa were analysed by FCM. Results MTT assay and Colon formation method showed CMNa had radiosensitive effect on hypoxic laryngeal carcinoma Hep-2 cell line. SER was 1.23,1.54,1.98. FCM analysis showed Hep-2 cells treated with CMNa were accumulated in G2/M phases. Higher the concentration,higher the percentage of G2/M phase. Conclusion CMNa has obvious radiosensitive effect on hypoxic laryngeal carcinoma Hep-2 cell line, the effect increases along with the increase of concentration. One of the radiosensitiv- ity mechanisms of CMNa may be that it can decrease the proportion of the hypoxic laryngeal carcinoma Hep-2 cells in the G0/G1 phase which are with radiation tolerance, and increase the proportion of G2/M phase that cells are radiosensitive.
