基于SELDI-TOF-MS技术的慢性肾衰竭4种舌苔相关蛋白研究

摘　　要：目的:采用表面增强激光解析离子化飞行时间质谱技术(SELDI-TOF-MS)技术研究慢性肾衰(CRF)患者4种类型舌苔中的差异蛋白标志物。方法:采集CRF患者的舌苔102例,其中腻苔组56例(白腻苔组24例、黄腻苔组32例)、非腻苔组46例,正常对照组的薄白苔38例,采用NP20蛋白芯片技术进行舌苔蛋白质组学研究,用蛋白芯片阅读器PBSⅡ对芯片进行扫描、分析。结果:在质荷比1 000-20 000范围内,腻苔组与正常对照组有17个差异蛋白峰(P<0.01),非腻苔组与正常对照组有19个差异蛋白峰(P<0.01),腻苔组与非腻苔组有9个差异蛋白峰(P<0.01),白腻苔组与正常对照组有9个差异蛋白峰(P<0.01),黄腻苔组与正常对照组有11个差异蛋白峰(P<0.01)。结论:初步筛选出了CRF 4种类型的舌苔蛋白标志物,为CRF中医舌诊的客观化研究提供了一定的实验依据。Objective： To investigate the protein markers in four kinds of tongue coating in patients with chronic renal failure（CRF） based on SELDI-TOF-MS technique.Methods： The tongue coating was taken from 102 patients with CRF and 38 healthy people,there were 56 cases greasy tongue,46 cases non-greasy tongue among the 102 patients.Then the tongue coating was investigated by proteomic techniques with NP20 gene chip.PBSII was used to scan and analyse chip.Results： In the greasy tongue coating group and control group,17 different protein peaks were identified in the protein map（P0.01）.In the non-greasy tongue coating group and control group,19 different protein peaks were identified in the protein map（P0.01）.In the greasy tongue coating group and non-greasy tongue coating group,9 different protein peaks were identified in the protein map（P0.01）.In the white-greasy tongue coating group and control group,9 different protein peaks were identified in the protein map（P0.01）.In the yellow-greasy tongue coating group and control group,11 different protein peaks were identified in the protein map（P0.01）.Conclusion： This research screenes out 4 kinds of tongue coating protein markers,which provides experimental evidence for objective of the tongue study of TCM in CRF.
