黑素细胞中GPNMB与Tyr相互作用的探讨

摘  要:目的:探讨非转移性黑色素瘤糖蛋白B(glycoprotein(transmembrane)nonmetastatic melanomaprotein b,GPNMB)与酪氨酸酶(tyrosinase)的相互作用。方法:以黑素细胞系PIG1为研究对象,分别构建GPNMB和Tyr的siRNA以及过表达载体,转染入细胞后采用实时定量RT-PCR、Western blot检测GPNMB表达下调和上调对Tyr的影响,以及Tyr表达下调和上调对GPNMB的影响;采用免疫共沉淀法检测它们之间的直接相互作用。结果:GPNMB表达下调和上调对Tyr有影响,而Tyr表达下调和上调对GPNMB几乎没有影响。结论:黑素细胞中GPNMB与Tyr之间不存在直接相互作用,为进一步研究GPNMB在黑素小体形成中的作用机制奠定了基础。Objective To explore the interaction between glycoprotein (transmembrane) nonmetastatic melanoma protein b (GPNMB) and tyrosinase. Methods siRNA and overexpression vectors of GPNMB and Tyr were constructed, respectively, and transfected into PIG1 melanocytes. Real-time quantitative RT-PCR,Western blot were used to determine the effect of GPNMB down-regulation and up-regulation on Tyr,as well as the effect of Tyr down-regulation and up-regulation on GPNMB. The direct interaction between GPNMB and Tyr was detected by the method of immune-precipitation. Results Tyr expression was influenced by GPNMB down-regulation and up-regulation, whereas Tyr down-regulation and up-regulation has almost no effect on GPNMB. Conclusion There was no direct interaction between GPNMB and Tyr in PIG1 melanocytes, which laid the foundation for studying the mechanism of GPNMB in the formation of melanosomes.
