摘　　要：目的探讨雌激素对人正常皮肤成纤维细胞骨形态发生蛋白-2(BMP-2)表达的影响。方法采集正常人皮肤标本,分离培养真皮成纤维细胞。分别在培养细胞中加入PBS、雌激素、BMP-2刺激,应用流式细胞仪观察细胞周期变化情况,通过反转录-聚合酶链反应(RT-PCR)法观察BMP-2mRNA表达情况。结果雌激素和BMP-2处理组细胞增生指数与对照组比较差异具有统计学意义(P<0.01)。雌激素处理组BMP-2mRNA表达显著高于对照组(P<0.05)。结论雌激素可上调人正常皮肤成纤维细胞BMP-2mRNA的表达,促进成纤维细胞增生。Objective To investigate the effects of estrogen on bone morphogenetic protein-2 gene transcription in human skin fibroblasts. Methods Human dermal fibroblasts were isolated by trypsin digestion method from normal adult skin specimens and cultured in DMEM containing 10% newborn bovine serum. The cellular periodicity distribution was detected by flow cytometry after stimulated by PBS, estrogen and BMP-2 protein respectively. The change of BMP-2 mRNA expression was detected by RT-PCR after stimulated by estrogen. Results The proliferation ratio of cells stimulated by estrogen and BMP-2 protein was higher than the control group （P 〈 0.01）. The expression of BMP-2 mRNA increased significantly in 17-β-estradiol treatment group. Conclusion 17-β-estradiol treatment could upregulate BMP-2 mRNA expression and promote cell proliferation.
