强筋健骨胶囊对急性痛风模型炎症因子的影响

摘　　要：目的探讨强筋健骨胶囊治疗急性痛风性关节炎的作用机制。方法实验大鼠右后足踝关节腔注射尿酸钠溶液建立急性痛风性关节炎模型,观察右后足踝关节肿胀程度,计算关节肿胀率;实验结束时,颈总动脉采血,分离血清,酶联免疫吸附测定法(ELISA)测定血清白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、肿瘤坏死因子(TNF-α)和基质金属蛋白酶3(MMP-3)的含量。结果强筋健骨胶囊显著降低模型大鼠关节肿胀度(P<0.05);显著降低血清TNF-α、IL-1β、IL-6及MMP-3的水平(P<0.05)。结论强筋健骨胶囊通过降低IL-1β的水平,减少刺激单核-巨噬细胞合成IL-6、TNF-α,降低其诱导产生MMP-3,从而减轻炎症反应,这可能是强筋健骨胶囊治疗痛风性关节炎急性发作期的作用机制。Objective To investigate the effect and mechanism of Qiangjinjiangu Capsules on acute gouty arthritis.Method An acute gouty arthritis model was established by injection of uric acid sodium solution to test rats′ right rear ankle joint cavity.The swelling degree of the right rear ankle joint was observed and calculated.The IL-6,IL-1β,TNF-α,and MMP-3 of the serum,isolated from the common carotid artery,was determined by ELISA.Result The joint swelling degree of Qiangjinjiangu Capsules group decreased obviously（P0.05）;TNF-α,IL-1β,IL-6α and MMP-3 in serum were reduced significantly（P0.05）.Conclusion Qiangjinjiangu Capsules showed effects in the treatment of acute gouty arthritis.Its mechanism involves the lowering IL-1β,decreasing excite monocyte-macrophages compound IL-6 and TNF-α,and reducing MMP-3.
