摘 要:目的探讨亚硝酸盐对邻苯三酚红钼酸盐法尿液总蛋白检测结果的影响。方法收集不同尿总蛋白浓度的新鲜尿液标本80份,每份标本分成6管,每管1ml,取1管作为对照,其余分别加入不同含量的亚硝酸钠,终浓度依次50、100、200、500和1000μmol/L。用邻苯三酚红-钼酸盐法检测,80份标本分成4个水平(各20份)进行统计分析。结果亚硝酸钠含量越高影响越大,尿液中总蛋白浓度越低,对检测影响也越大。亚硝酸盐含量在200μmol/L以上开始出现部分结果差异有统计学意义(P﹤0.01),500μmol/L亚硝酸钠使156mg/L和327mg/L尿总蛋白下降达到14.5%和12.5%,1000μmol/L亚硝酸钠使642mg/L尿总蛋白下降达到17.7%。结论亚硝酸钠对邻苯三酚红-钼酸盐法尿液总蛋白检测结果存在负干扰。OBJECTIVE To investigate the effect of nitrite on urine total protein measurement with the pyrogallol red-molybdate method. METHODS Fresh urine specimens were collected from 80 out patients, each divided into six aliquots, one group was chosen as control, the others were added to different levels of sodium nitrite, the final concentrations of sodium nitrate were 50, 100, 200, 500 and 1 000μmol/L respectively. Urine total protein concentrations were analyzed by the pyrogallol redmolybdate method, and 80 samples were divided into four levels for statistical analysis. RESULTS The urine total protein decreased when the concentration of sodium nitrite increased. The urine total protein concentrations decreased to 14.5% and 12.5% when 500μmol/L of sodium nitrite were added into urine at 156mg/L and 327mg/L of the urine total protein, and decreased to 17.7% with 1 000μmol/L of sodium nitrite at 642 mg/L of the urine total protein. CONCLUSION The sodium nitrite could affect the urine total protein assay with the pyrogallol red-molybdate method and cause a false negative resuh.
