摘 要:【目的】寡糖可以诱导毛白杨愈伤组织产生防卫反应,以提高其抗病性,从而抵御杨树溃疡病菌的入侵。【方法】分别采用筛选出的杨树溃疡病菌菌丝体、壳聚糖、果胶糖活性较高的降解物寡糖A1、B、C3诱导毛白杨愈伤组织,以加无菌水为对照(CK),并在诱导48h后用针刺接种法接种杨树溃疡病菌,检测毛白杨愈伤组织中丙二醛(MDA)、富含羟脯氨酸糖蛋白(HRGP)及绿原酸含量的动态变化。【结果】3种寡糖A1、B、C3分别诱导48h后接种杨树溃疡病菌,毛白杨愈伤组织中MDA含量变化先略有升高,之后缓慢下降,在接种后60h急剧升高,并在接种60~96h毛白杨愈伤组织中MDA含量变化为:CK>寡糖C3>寡糖B>寡糖A1;毛白杨愈伤组织中HRGP含量在0~96h均有不同程度的增加,并且寡糖A1诱导并接种的毛白杨愈伤组织中HRGP和绿原酸含量明显高于寡糖B、C3及对照,毛白杨愈伤组织中绿原酸含量的变化趋势呈单峰曲线。方差分析表明,与接种前相比,3种寡糖A1、B、C3分别诱导并接种后毛白杨愈伤组织中MDA、HRGP及绿原酸含量均有极显著增加(P<0.01)。【结论】寡糖诱导并接种杨树溃疡病菌后,毛白杨愈伤组织中的MDA、HRGP及绿原酸含量均有明显提高,从而增强其抗病性。【Objective】 Oligosaccharides induced defense response improves disease resistance of the Populus tomentosa callus and resists the infection of Dothiorella gregaria.【Method】 The P.tomentosa calluses were induced by oligosaccharide A1,B and C3 which had high activity and were respectively selected through the degradation of D.gregaria,chitosan and protopecten.Aseptic water was used as the CK.First the P.tomentosa calluses were induced by oligosaccharides for 48 hours,then the inducted and CK calluses were inoculated by D.gregaria.The dynamic changes of MDA,HRGP and chlorogenic acid in P.tomentosa calluses were assayed.【Result】 The contents of MDA in P.tomentosa calluses inoculated by D.gregaria after being induced by oligosaccharide A1,B and C3 for 48 hours increased initially and reached the first peak.Then they decreased slowly until sharply raised after 60 h.The contents of MDA in P.tomentosa calluses induced by oligosaccharide A1 was the lowest in the four treated calluses(60-96 h).The contents of HRGP in four treatments increased at different degrees(0-96 h).The contents of HRGP and chlorogenic acid in calluses inoculated with D.gregaria after induced by oligosaccharides A1,were higher than that of other treatments.The contents of chlorogenic acid in all treatments showed a single peak curve.Variable analysis indicated that the variations of contents of MDA,HRGP and chlorogenic acid of calluses inoculated by D.gregaria after being induced by oligosaccharides A1,B and C3 showed highly significant differences(P0.01).【Conclusion】 In certain low concentration range,the contents of MDA,HRGP and chlorogenic acid were improved significantly in the P.tomentosa calluses induced by oligosaccharides.The oligosaccharides can enhance the disease resistance of poplar celluses.
