摘　　要：目的 :建立高效液相色谱法测定人血清中醋酸甲地孕酮浓度。方法 :分析柱为YWG -C1 8(10 μm ,4.6mm× 2 5 0mm) ;流动相为水 -乙腈 (40∶6 0 ) ;流速为 1.0ml min ;紫外检测波长为 2 88nm ;用乙酸乙酯作为提取液。结果 :血清中醋酸甲地孕酮浓度在1～ 12 8ng·ml- 1 范围内线性关系良好 (r =0 .9997) ;醋酸甲地孕酮高、中、低 3个浓度的日内、日间变异系数分别小于 10 %和20 % ;平均回收率为 98.5 5 % ;最低检测血药浓度为 0 .5ng·ml- 1 。结论 :本方法准确、方便 ,灵敏度高 ,适合用于临床血药浓度监测和药代动力学研究。Aim The serum concentration of megestrol acetate was determined with RP-HPLC method.Methods After being extracted with ethyl acetate,megestrol acetate was analyzed by RP-HPLC (YWG-C 18 )and UV-detector (288nm).The mobile phase was a mixture of acetonitrile and water(60∶40) at a rate of 1.0ml/min.Results The linear range was 1～128ng·ml -1 (r=0.999 7).The average recoveries of megestrol acetate was 98.55%.RSD of intra- and inter-days was less than 10% and 20% respectively.Conclusion This method possesses high accuracy and precision.It is sensitive and specific.
