摘  要:目的:建立同时测定栀子中去乙酰车叶草苷酸甲酯、栀子苷含量的HPLC分析方法。方法:色谱柱:Agilent Eclipse XDB-C_18(4.6mm×250mm,5μm);流动相:乙腈-0.1%磷酸水溶液,梯度洗脱。结果:去乙酰车叶草苷酸甲酯、栀子苷分别在0.009~0.186mg/mL(r=0.9994)、0.042~0.846mg/mL(r=0.9996)浓度范围内与峰面积呈良好的线性关系,平均回收率分别为97.08%,RSD=1.48%和100.45%,RSD=1.29%(n=6)。结论:所建立方法简单、准确,易操作,具有良好的精密度、重复性和回收率,可作为栀子中这两种成分的含量测定方法。Objective:To establish a method using HPLC for the simultaneous content determination of geniposide and deacetyl asperulosidic methyl ester in Fructus Gardeniae. Methods:The HPLC method was applied with Agilent Eclipse XDB-C18 column, The mobile phases consisted of acetonitrile and 0.1% phosphoric acid aqueous solution, gradient elution, the detection wave length was at 238 nm and the flow rate was 1ml/min. Results: Deacetyl asperulosidic methyl ester and geniposide showed a good linear relationship in the range of 0.009-0.186mg/ml(r=0.9994),0.042-0.846mg/ml(r=0.9996) respectively, the average recovery were 97.08% and 100.45% with RSD at 1.48% and1.29% (n=6). Conclusion: The method is simple, accurate and easy to operate, has good precision, repeatability and recovery. It can be used to determine contents of these two ingredients in Fructus Gardeniae.
