脂质体免疫检测法测定血清总补体活性临床价值的探讨

摘　　要：目的：引进日本WAKO 公司应用脂质体免疫检测法定血清总补体活性，代替传统的总补体活性测定法（CH50）， 而不需要绵羊红细胞。方法：取血清10ul 与250ul 脂质体（试剂1） 混合，5min 后加入抗体基质（试剂2） 延迟5min 于340nm 波长下测定NADH 的吸光度，查标准曲线。结果：健康组40 名成人总补体活性为（X 2S ）40.9 13.1u/ml。 范围：27.8 54.1u/ml。 疾病组：减低组为37 例总补体活性低于24u/ml 以下。增高组为19 例总补体活性高于70u/ml 以上。结论：该方法最大优点是不用绵羊红细胞，血清用量少，操作简单10min 即可出结果，影响因素小，临床应用价值大，是一个值得推广的方法。o b j e t i v e:Import the immune detection method with liposome from WAKO company of Japan to measure the activity of serum total complemenl replacing the total complement- hemolysis-activity(ch50)measurement,without the use of SRBC.M e t h o ds:fetch 10ul blood serum and mix it with 250ul liposome (reagent1),5mins later,add the antibody (reagent2)to it ,keep it for 5mins,then measure the absorbance of NADH with wavelength being 340nm ,look into standard curve.R e s u l t s:health group(40adults):the activity of total complement is X S 40.9 3.1u/ml;range:27.8-54.1u/ml.Disease group reduction group(37adults)the activ- ity of total complement is lower than 24u/ml.Enhancement group(19)the activity of total complement is higher than 70u/ml.C o n c l u s i on:The greatest advantage is avoiding the use of SRBC,the amount of serum used is small ,the operation sequence is simple,the results can be achieved only in 10mins.inflnuential factors are rare.The value of clinineal application is relatively larger.It is a method worth popularizing.
