芍药提取物对老年自发性高血压大鼠血压、血脂以及肝功能的影响

摘　　要：[目的]观察芍药提取物对老年自发性高血压大鼠(SHR)血压、血脂以及肝功能的影响。[方法]15月龄SHR50只,随机分为模型对照组、卡托普利组(15 mg.kg-1)以及芍药提取物组(12.5 mg.kg-1、25 mg.kg-1、50 mg.kg-1);另取10只WKY大鼠作为正常对照组。单次给药,应用无创血压测量仪检测不同时间段大鼠收缩压(SBP)、舒张压(DBP)、平均动脉压(MBP)变化;连续给药4周,每周平行测量大鼠收缩压、舒张压、平均动脉压;末次给药后眼眶取血,测定血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-c)、低密度脂蛋白(LDL-c)、血清丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)水平。[结果](1)单次给药时,芍药提取物(12.5 mg.kg-1、25 mg.kg-1)能短暂地降低SHR舒张压、平均动脉压;芍药提取物(50 mg.kg-1)能显著降低SHR收缩压、舒张压、平均动脉压,降压作用维持到6 h。(2)连续给药期间,芍药提取物(50 mg.kg-1)能显著降低SHR收缩压、舒张压、平均动脉压。(3)芍药提取物各剂量能不同程度降低血清TC、TG、LDL-c水平。芍药提取物(50 mg.kg-1)还能降低血清ALT、AST水平。[结论]芍药提取物具有降血压以及调节血脂的作用,对肝脏功能具有一定的保护作用。[Objective] To study the effects of Herbaceous Peony extract on blood pressure,blood lipid levels and hepatic function of aged SHR.[Methods] 15 months old SHR were randomly divided into model control group,captopril group（15 mg·kg-1） and Herbaceous Peony extract group（12.5 mg·kg-1,25 mg·kg-1,50 mg·kg-1）.WKY was used as normal control group.After a single dose administration,blood pressure of different periods was measured by noninvasive blood pressure measurement instrument.After that,blood pressure was measured once a week during the period of successive administration.After the last administration,blood sample was drawn from orbit to detect serum triglyceride（TG）,total cholesterol（TC）,high-density lipoprotein（HDL-c）,low-density lipoprotein（LDL-c）,alanine aminotransferase（ALT）,aspartate aminotransferase（AST）.[Result]（1） After a single dose administration of Herbaceous Peony extract（12.5 mg·kg-1,25 mg·kg-1）,blood pressure of SHR decreased for a short period of time.Herbaceous Peony extract（50 mg·kg-1） could also decrease the blood pressure and the antihypertensive effect lasted for six hours.（2） After successive administration for four weeks,blood pressure of SHR in Peony extract（50 mg·kg-1） group decreased significantly.（3） All dosage of Herbaceous Peony extract could decrease serum TC,TG,LDL-c levels.Herbaceous Peony extract（50 mg·kg-1） could also significantly decrease serum ALT,AST levels.[Conclusion] Herbaceous Peony extract could decrease blood pressure,regulate blood lipid and protect the liver function of aged SHR.
