摘 要:为寻找GH基因编码区存在突变位点及突变对太湖鹅10周龄体重关联性,试验在太湖鹅生长激素基因第2、3、4、5外显子上各设计1对引物,用SSCP方法寻找存在点突变,分析10周龄体重在不同基因型间差异。结果表明,在第2外显子中发现T39C、T74C突变,T74C突变导致氨基酸V25A处发生有义突变。在外显子4中发现T291C、G297A突变,均未引起氨基酸变化。不同基因型频率在太湖鹅群体中差异较大。太湖鹅群体平均纯合度为0.4913,杂合度为0.5087,多态信息含量为0.4108。10周龄太湖鹅体重在不同基因型间均无显著性差异,但外显子2上突变对体重影响在公母鹅中表现一致。GH基因在太湖鹅群体中遗传多样性较为丰富,初步探讨GH基因外显子突变对太湖鹅10周龄体重影响。The purpose was to find mutational site in coding region of GH gene, and the relevance between mutation and 10-week-old body weight of Taihu goose. In this experiment, four pairs of primers were designed to amplify the exon 2, 3, 4, 5 of GH gene in Taihu goose, and then single nucleotide polymorphisms (SNPs) were detected by the technique of single strand conformation polymorphism (SSCP) and the body weight was compared among different genotypes. Two nucleotide variations including T39C, T74C were found in exon 2, and the second variation resulted in the amino acid variations at V25A. Two nucleotide variations includingT291C, G297A were found in exon 4 and they don't result in the amino acid variations. The population genetics indicated that homozygosity, heterozygosity and polymorphic information content (PIC) was 0.4913, 0.5087, 0.4108 respectively. The body weight of 10-week-old Taihu goose don't have significant difference among different genotypes. But the relation of variations of exon 2 and body-weitht was the same in sex. The GH gene had high polymorphisms in Taihu goose breed. The influence has been first explored between mutation of exons in GH gene and 10-week-old body weight.
