柴胡复方水提物对大鼠急性胃溃疡预防作用的影响

摘　　要：目的:研究柴胡复方水提物对大鼠急性胃溃疡预防作用的影响。方法:大鼠灌胃给予消炎痛(70 mg/kg)复制急性胃溃疡模型,测定胃溃疡指数和溃疡抑制率,常规苏木素-伊红染色观察胃黏膜病理形态学的改变,一氧化氮硝酸还原酶法测定血清一氧化氮(NO)含量的变化,黄嘌呤氧化酶法测定血清超氧化物歧化酶(SOD)的活力,观察柴胡复方水提物对大鼠急性胃溃疡的预防作用。结果:阳性药物Stillen组(100 mg/kg)、柴胡复方水提物低剂量(100 mg/kg)和高剂量组(200 mg/kg),溃疡指数明显低于溃疡模型组(P<0.01),溃疡抑制率分别为48%、79%和80.9%,血清SOD较模型组显著降低(P<0.01);与模型组比较,柴胡复方水提物高低剂量组血清NO降低(P<0.05);胃组织病理形态学观察,模型组胃黏膜层大量缺失,严重者已脱落坏死,黏膜下层充血、水肿,且有少量纤维素、炎性细胞浸润,阳性药物组和柴胡复方高低剂量组胃组织病变有明显改善。结论:柴胡复方水提物对大鼠急性胃溃疡模型有明显的预防保护作用。Objective： To study effects of radix bupleuri compound water extraction on prevention in rats with acute gastric ulcer.Methods： The model of acute gastric ulcer was established by indomethacin gavage（70mg/kg） to observe effects of radix bupleuri compound water extraction on prevention in rats with acute gastric ulcer.The ulcer index and inhibition ratio of ulcer were measured.The slices of gastric tissues were observed after HE staining.Nitric oxide（NO） and superoxide dismutase（SOD） in serum were measured by nitric oxide nitric oxide reductase assay and xanthine oxidase assay.Results： In Stillen group（100 mg/kg）,low dose of radix bupleuri compound water extraction group（100 mg/kg） and high dose group（200 mg/kg）,the ulcer indexs were decreased compared with those in model group（P0.01）.The ulcer inhibition ratios were 48 %,79 % and 80.9 %,and the activities of SOD in serum were decreased in drug-administrated groups（P0.01）.The content of NO of serum in radix bupleuri compound water extraction groups were diseased compared with those in model group（P0.05）.In model group,desquamated gastric mucosas were observed and necrosis in serious animals,hyperemia and edema with inflammatory cells infiltration in submucosa.Morphological changes were improved in drugs-administrated groups.Conclusions： Radix bupleuri compound water extraction can prevent for in rats with acute gastric ulcer.
