摘 要:目的探讨低强度脉冲超声(low intensity pulsed ultrasound,LIPUS)对人牙周膜细胞(human periodontal ligament cells,hPDLCs)增殖和碱性磷酸酶(alkali phosphatase,ALP)活性的影响。方法不同参数(按辐照强度I_(SATA) 30、60、90 mW/cm^2,时间10、20、30 min/d分组)LIPUS辐照干预hPDLCs培养,四甲基偶氮唑盐比色法检测hPDLCs增殖率,流式细胞仪检测细胞增殖周期,酶化学法测定ALP活性。结果 LIPUS辐照时间20 min/d、强度90 mW/cm^2时可促进hPDLCs增殖;辐照后hPDLCs增殖指数较对照组显著提高。各组ALP活性较对照组均有不同程度提高;不同辐照强度组间及不同辐照时间组间的ALP活性差异显著(Fi=226.391,P<0.05,Ft=145.572,P<0.05);不同辐照强度和辐照时间的交互作用也有统计学意义(F=448.540,P<0.05);其中90 mW/cm^2、20 min/d组促进作用最强。结论 LIPUS具有促进hPDLCs增殖及成骨分化作用,90 mW/cm^2、20 min/d为促进hPDLCs增殖和成骨分化的适宜参数。Objective To explore the effect of low intensity pulsed ultrasound (LIPUS) on proliferation and alka li phosphatase (ALP) activities of human periodontal ligament cells (hPDLCs). Methods hPDLCs were isolated by tissue mass method and evaluated by immunohistochemistry, hPDLCs cultured in vitro were irradiated by LIPUS of various dosage (intensity of 30, 60, 90 mW/cm2 ,time of 10, 20, 30min/d). Cell proliferation was determined by MTT method, cell cycle was assayed by flow cytometry, ALP activity was examined by enzyme-chemistry. Results LIPUS of I_SATA 90 mW/cm2 and 20 min/d groups could improve cell proliferation. Proliferation index of hPDLCs was increased significantly by LIPUS irradiation and the variation trends was consistent with control group. ALP activity of each experimental group was increased to different extent. Difference between the various irradiation intensity groups and various irradiation time groups was significantly(F/ 226. 391, P〈0.05, Ft= 145. 572, P〈0.05), there are statistical significance of interaction between various irradiation intensity and time effect(F=448. 540, P〈0.05) and 90 mW/cm2 -20 min/d group showed the strongest effect among all the experimental groups. Conclusions LI PUS could simulate hPDLCs proliferation and osteogenisis, 90 mW/cm2 -20 min/d may be the proper dosage in this study.
