摘 要:目的建立小鼠放射性肺损伤模型。方法 80只雌性C57BL/6小鼠随机分为四组:A、B和C三组每组24只小鼠,分别单次X线照射12、20和30Gy;D组8只小鼠作为空白对照。照射后1、3d和1、2、4、8、16、24周8个时点取出肺组织,行HE、VG染色,观察肺纤维化形成,RT-PCR测定肺组织转化生长因子(TGF)-β1的表达。结果 A、B、C组均出现典型放射性肺损伤及肺纤维化表现,随着照射剂量的增加,肺部病变加重,且肺部纤维化出现提前。与D组相比,A、B、C组8个时点肺组织TGF-β1含量均显著增高(P<0.01)。结论成功建立了放射性肺损伤小鼠动物模型。Objective To establish a mouse model with radiation-induced lung injury. Methods Eighty female mice were randomly divided into groups of A(12 Gy, 24 mice), B(20 Gy, 24 mice), C(30 Gy, 24 mice) and D(controls, 8 mice). At eight time points of 1,3 d, and 1,2,4,8,16,24 weeks after irradiation, the lung tissues were observed by HE and VG staining and the expressions of transforming growth factor(TGF)-βl in the lungs were detected by RT-PCR Results The typical radiation-induced lung injuries and pulmonary fibrosis were seen in groups of A, B and C. As the increase of irradiation dosage, the lung lesions aggravated and pulmonary fibrosis appeared in advance. Compared with group D, the expressions of TGF-βl in groups of A,B and C were significantly higher at all time points(P〈0. 01). Conclusion The mouse model of radiation-induced lung injury has been successfully established.
