摘 要:目的研究不同细胞制备方法对细胞因子诱导的杀伤细胞(CIK)细胞亚群比例的影响。方法将42份CIK细胞随机均分为实验组和对照组。对照组按常规CIK细胞制备方法制备;实验组在对照组基础上于第2天加入重组人粒细胞集落刺激因子(rhG-CSF)10ng/ml。待细胞成熟后采用流式细胞术检测两组CD3+CD4+、CD3+CD8+和CD3+CD56+T淋巴细胞比例的变化。结果与对照组相比,实验组CD3+CD4+T细胞比例下降(P<0.01),CD3+CD56+T细胞比例升高(P<0.05);CD3+CD8+T细胞比例稍高,但无统计学差异(P>0.05)。结论在CIK细胞制备过程中加入rhG-CSF可抑制CD3+CD4+T细胞,增加CD3+CD56+T细胞生成。Objective To study the effects of different cell preparation methods on proportion of cytokine-induced killer(CIK) cell subsets. Methods Forty-two CIK cell samples were equally randomized into two groups. Group A was prepared according to the conventional method and group B was treated with recombinant human granulocyte colony-stimulating factor(rhG-CSF) 10 ng/ml on the 2nd day on the basis of conventional method. Flow cytometry was used to detect the proportion of CIK cell subsets after cell maturation in two groups. Results Compared with group A, the proportion of CD3+ CD4^+ T cell was decreased(P〈0. 01), which of CD3^+ CD56^+ T cell was increased(P〈0. 05) in group B. There was no significant difference in the proportion of CD3^+ CD8^+ T cell between two groups(P〉0. 05). Conclusion rhG-CSF may inhibit the induction of CD3^+ CD4^+ T cells and improve the induction of CD3^+ CD56^+ T cells in the preparation of CIK cells.
