摘  要:目的:了解东莞市2011年流行的麻疹病毒的基因型别和核蛋白基因变异情况。方法:采集病人的咽拭子或尿液标本,用VERO-SLAM细胞分离病毒。通过逆转录-聚合酶链反应(RT-PCR)和测序技术获得毒株的核蛋白基因碳末端450 bp核苷酸序列,并与麻疹各基因型代表株及沪191疫苗株进行序列比对和构建进化树分析。结果:共分离到6株麻疹病毒株,均属于H1基因型H1a亚型。6株毒株间的核苷酸同源性为99.8%~100%。6株毒株间有5株序列相同。各毒株与China93-2毒株同源性最高,为98.7%和98.9%。与沪191疫苗株同源性则为92.4%~92.7%。结论:东莞市2011年麻疹病毒流行株均属于H1基因型H1a亚型,与中国的流行情况一致。暂无新型及传入性的其它亚型出现。毒株间的变异很小。Objective: To know the genotype of measles viruses circulating in 2011 in Dongguan city and the mutation situation of the nucleoprotein gene. Methods: The throat swabs and urine specimens from measles patients were collected for viruses isolation by inoculating the specimens to Vero - Slam cell line. The 450 bp COOH -termi- of nucleoprotein gene fragment was obtained by RT - PCR and sequencing analyzed by sequence alignment and reconstructing phylogenetic tree. Results technology. The target fragment was : Total 6 viral strains were isolated from patients. Phylogenetic analysis of the N gene sequences showed that all of the isolates belonged to H1 genotype H1 a subtype. The homology of nucleotides sequence of these viruses ranged from 99.8% - 100%. Five virus strains had the same sequence. The viruses had 98.7% ~ 98.9% and 92.4% -92.7% homology of nucleotides sequence with China93_2 virus and shanghail91 vaccine strain virus respectively. Conclusion: All 6 measles viral strains cir- culating in Dondguan city are H1 genotype Hla subtype, it is in consistency with the situation of China. No other genotype or new genotype emerged in Dongguan city. The genetic diversity of HA gene are low among these virus strains
