摘 要:目的:建立牛奶蛋白质的快速光度分析方法。方法:将牛奶与1+1硫酸混合置于电压力锅内,加热加压水解40 min,水解产生的氨基酸在Sn2+催化下,与茚三酮反应显色后,分光光度法测定。结果:牛奶中蛋白质含量在3 mg~18 mg之间时与吸光度成线性,r=0.9992,最低检测量0.5 mg,方法回收率98.3%~102%,相对标准偏差2.45%~3.98%。结论:建立的方法适用于牛奶中蛋白质的快速测定。Objective: To establish a method for fast determination of protein in milk by spectrophotometry. Methods: After the milk was hydrolysed with H2 SO4 ( 1 + 1 ) under heated and pressurized condition for 40 min, the amino acids decomposed from protein reacted with ninhydrin to form colored products under catalysis of Sn^2+ and the colored products were determined by spectrophotometry. Results: The linear range of the method was 3 mg -18 mg (r = 0. 9992) with the detection limit of 0.5 mg and the recoveries of 98.3 % -102%, and the RSDs were 2.45% 3.98%. Conclusion : The developed method was suitable for the fast determination of protein in milk.
