摘  要:目的研究染料木黄酮对大剂量X射线辐射诱导的人脐静脉血管内皮细胞(HUVEC)氧化性损伤的防护作用。方法染料木黄酮浓度为0、5、20、50μmol/L分别于辐照前2 h预作用于HUVEC,以3.6Gy、6.0 Gy X射线照射后于24、48 h检测细胞内SOD、GSH-Px和ROS的水平变化。结果与正常对照组比较,辐照模型组HUVEC细胞内SOD、GSH-Px的含量明显降低(P<0.01),ROS含量显著增加(P<0.01),且随着辐照剂量的增加变化越明显;与辐照模型组相比,染料木黄酮给药组能明显改善辐射诱导的氧化应激引起的损伤,显著降低ROS含量,增加SOD活性(P<0.01),在一定浓度范围内,防护作用随着染料木黄酮浓度的增加而增强,且染料木黄酮对3.6 Gy辐照剂量下的防护效果优于6.0 Gy辐照剂量。与辐照模型组相比,染料木黄酮各组细胞内GSH-Px水平无显著变化(P>0.05)。结论染料木黄酮有一定的抗辐射作用,在一定浓度范围内其防护效果呈现浓度依赖性。Objective To study the protective role of genistein on high dose X-radiation-induced oxidative stress damage in HUVECs.Methods Before exposing to 3.6 Gy or 6.0 Gy X-ray,HUVECs incubated with different concentrations of genistein for 2 h.After 24 h and 48 h incubation,the levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and reactive oxygen species(ROS) were detected.Results Compared with the control group,the levels of ROS were apparently increased(P〈0.01),but SOD and GSH-Px were significantly decreased(P〈0.01) in the X-radiation-treated group.Compared with the X-radiation-treated group,ROS levels were markedly decreased(P〈0.01),SOD levels were obviously increased,but GSH-Px levels did not show a statistical difference in genistein-treated group(P〈0.05).The protection of genistein against 3.6 Gy X-ray irradiation was superior to that of 6.0 Gy X-ray irradiation.Conclusion Genistein can ameliorate X-radiation-induced oxidative damage in HUVECs at a dosage dependent manner.
