摘 要:目的研究重组人促红细胞生成素对新生鼠坏死性小肠结肠炎的保护作用。方法 30只新生1日龄大鼠随机分成三组:EPO干预组(A)、模型组(B)及正常组(C)。A组腹腔注射EPO 300μ/kg.d,隔日一次。B组及C组未予任何药物。第8天A及B组行缺氧复氧冷刺激1次/日,连续2天,建立新生鼠NEC模型。所有动物于造模后24小时断头处死,取近回盲部近端肠管1~2 cm固定包埋,切片、HE染色,剩余肠组织制备组织匀浆,取上清液检测PAF、IL-8、IL-10的含量。结果 A组肠组织病理切片显示较B组肠壁损伤轻,组织病理学评分低(P<0.05),肠组织PAF、IL-8分别为(18968.78±2548.65 pg/g.pro,40.65±9.24 ng/g.pro),均低于模型组(P<0.05),而IL-10为74.34±5.27 pg/g.pro,高于模型组(P<0.05)。结论重组人促红细胞生成素预处理可以减轻新生鼠NEC的肠道炎症反应,对新生鼠坏死性小肠结肠炎有保护作用。Objective To observe effect of recombinant human erythropoietin(rhEPO) on neonatal rats with necrotizing enterocolitis.Methods Thirty one-day-old SD rat pups were randomly divided into 3 groups: NEC+EPO group(A),NEC model group(B) and normal control group(C),each group had 10 rats.Group A were Intraperitoneal injection of rhEPO 300u/kg.d,once every other day,group B and C without any drug.From the eighth day,group A and group B were subjected to cold exposure shortly after hypoxic-reoxygenation treatment,once a day,for 2 consecutive days.All pups were killed 24h after NEC models making.The ileocecal segment of proximal segment of the colon were fixation,embedding,sectioning,staining for HE and histopathological examination.The remaining intestinal tissue were preparaed for tissue homogenate,which supernatant were detected for PAF,IL-8,IL-10.Results Group A intestinal histopathology showed bowel wall light injury,the organization pathology score was lower than that of group B(P0.05);The level of PAF,IL-8 were(18968.78±2548.65 pg/g.pro),(40.65±9.24 ng/g.pro),which less than of group B;the level of IL-10 was 74.34±5.27 pg/g.pro,which higher than that of group B(P0.05).Conclusions Pretreatment of rhEPO can reduce NEC in neonatal rats intestinal inflammatory response,hypoxia-induced intestinal injury and has protective effect on neonatal rats with necrotizing enterocolitis.
