摘 要:目的建立测定芎姜复方CO2超临界提取物中总内酯和总萜类含量的方法。方法以洋川芎内酯A为对照,30 mg.mL-1的氢氧化钠溶液,30℃恒温反应30 min,于305 nm波长处测定吸光度,测定总内酯含量;以6-姜酚为对照,5%香草醛-冰乙酸溶液、高氯酸显色,于560 nm波长处测定吸收度,测定总萜类含量。结果洋川芎内酯A在0.062 5~1.0 mg.mL-1与吸收度呈良好线性关系(r=0.999 9),平均回收率为97.9%,RSD为2.7%;6-姜酚在0.03~0.4 mg.mL-1与吸收度呈良好线性关系(r=0.999 9),平均回收率为97.6%,RSD为3.6%。结论本方法简便,准确,可用于芎姜CO2超临界提取物及其制剂中总内酯和总萜类含量的快速测定。Objective To establish a UV-Vis spectrophotometry method for the determination of total lactones and triterpenoids in CO2-SFE of Xiongjiang mixture.Methods senkyunolide A as the control,the total lactones were measured by UV-Vis spectrophotometry with 30 mg.mL-1 sodium hydroxide solution and in 30 ℃ insulation reacting for 30 min,the absorbance was measured at 305 nm wavelength.with 6-gingerol as the control,the total triterpenoids were measured by UV-Vis spectrophotometry with 5% vanillin-glacial acetic acid solution and perchloric acid as the color-developing agent,detected at 560 nm wavelength.Results Senkyunolide A had good linearity(r = 0.999 9) at 0.062 5-1.0 mg.mL-1,the average recovery was 97.9%,with RSD of 2.7%,while 6-gingerol had good linearity(r = 0.999 9) at 0.035-0.40 mg.mL-1,the average recovery was 97.6%,with RSD of 3.6%.Conclusion The method is simple,accurate and can be used for the determination of total lactones and triterpenoids in CO2-SFE of Xiongjiang mixture.
