摘  要:本文利用液相色谱-质谱/质谱(LC-MS/MS)联用检测技术,建立了一种新的、快速直接同时测定吸烟者血清中尼古丁及其9种代谢物的分析方法。各化合物的校准曲线用线性回归拟合且相关系数均大于0.99,回收率在75%~105%之间,定量限均低于0.85 ng/mL,方法的相对标准偏差在0.95%~10%之间。实验设计了70余名吸烟者摄入不同烟气量,并对其血液、尿液、唾液样本进行检测,以血清中所有代谢物含量做烟气摄入水平的分类分析,分类的正确度达到80%以上。各代谢物间的相关性及对分类的贡献度的研究表明,血清中的可的宁含量可作为不同烟气摄入量吸烟者的良好标记物。分析不同体液间可的宁的相关性,结果表明各体液间的可的宁含量显著相关。A novel validated,efficient liquid chromatography-tandem mass spectrometry(LC-MS/MS) procedure was developed and fully validated for the simultaneously determination of nicotine,cotinine,nicotine-N-oxide,nornicotine,norcotinine,cotinine-N-oxide,nicotine-N-β-D-glucuronide,trans-3-hydroxycotinine,cotinine-N-β-D-glucuronide,trans-3-hydroxycotinine-N-β-D-glucuronide in human serum.Calibration curves were linear over the selected concentration ranges for each analyte,with calculated coefficients of determination(R2) of greater than 0.99.The recovery of the method was ranged between 75-105%.The LOQ for all analytes were ≤0.85 ng/mL.Imprecision for the method(% R.S.D.) was ≤10%.Class modeling of 75 smokers' serum sample indicated the contents of all the metabolites in serum could approximately predict the status of smokers with different levels.The correlation analysis and contribution degree of these metabolites indicated that the cotinine in serum proved to be a good biomarker for valuation of different smoking level smokers.Furthermore,the correlation coefficient of cotinine content in serum and saliva embodied a distinct linearity of cotinine level in three body fluids.
