摘 要:目的观察芪地合剂含药血清对体外培养的大鼠肾小球系膜细胞(GMCs)增殖的影响,探讨其在糖尿病肾病(DN)防治中的可能机制。方法体外培养大鼠肾小球系膜细胞株,利用噻唑蓝(MTT)比色法动态观察各组含药血清作用下各组GMCs不同时间点的增殖情况。结果与正常组相比,高糖+LPS能明显刺激体外培养的GMCs增殖,而与模型组相比,芪地合剂含药血清能明显抑制GMCs增殖。结论高糖+LPS可促进GMCs增殖,芪地合剂含药血清能明显抑制高糖+LPS条件下GMCs增殖,从而发挥预防和治疗肾小球硬化,进而延缓DN的进展。Objective By observing the effects of drug-containing serum of Qidi mixture on glomerular mesangial cell (GMC) proliferationon in vitro under high glucose, significance in the prevention and treatment of diabetic nephropathy (DN) was explored. Methods The thiazolyl blue tetrazolium bromide (MTT) colorimetric method was used to observe the glomerular mesangial cell proliferations. Results Compared with the normal group, in vitro cultured glomerular mesangial cells were obviously proliferating under the stimulation of high glucose and LPS. Compared with the model group, drug-containing serum of Qidi mixture can inhibit GMC' s proliferatio indicated that there was no significant difference between the groups. Conclusion In vitro n, which cultured glomerular mesangial cells obviously proliferated under the stimulation of high glucose and LPS, but drug-containing serum of Qidi mixture could inhibit GMC' s proliferation which might be helpful in preventing the progression of diabetic nephropathy.
