摘 要:在碱性条件下,芦丁和维生素C对鲁米诺-铁氰化钾化学发光体系具有显著的抑制作用,但两者峰值出现的时间存在明显差异。基于这一特点,本文将偏最小二乘法与流动注射化学发光法结合,建立了偏最小二乘法-流动注射化学发光同时检测芦丁和维生素C的新方法。在优化实验条件下,发光强度ΔI与芦丁浓度和维生素C质量浓度在0.02~0.6 mg·L-1和0.1~20 mg·L-1范围内呈线性关系,检出限分别为4.5μg·mL-1和8.9ng·mL-1(信噪比为3)。经实际测定复方芦丁片和人体尿液中芦丁和维生素C的含量表明,该方法具有灵敏度高、分析速度快及操作简便等优点,适合用于药品及生物样品等复杂样品中芦丁和维生素C的测定。A flow-injection chemiluminescence(CL) system was developed for the simultaneous determination of ascorbic acid(AA) and rutin(RT) using partial least squares(PLS) calibration.This method is based on that fact that both AA and RT can quantitatively reduce Fe3+ to Fe2+,and that the reaction rates of AA and RT with Fe3+ are different.The calibration curve was linear over the concentration range of 0.1-20.0 mg·L-1 and 0.02-0.6 mg·L-1 for AA and RT,respectively.The limits of detection(LOD) for AA and RT were 8.9 μg·L-1 and 4.5 μg·L-1,respectively,which was calculated according to the International Union of Pure and Applied Chemistry(IUPAC) definition that is 3 times the standard deviation of the blank value.The proposed method was successfully applied to the simultaneous determination of both analytes in pharmaceutical formulations and human urine samples.The results indicate that the method offers the potential advantages of high sensitivity,simplicity and rapidity for AA and RT determination in pharmacy and biological samples.
