摘 要:目的观察局部应用微孔多聚糖微粒对创面愈合的影响并探讨其相关机制。方法建立C57BL/6J小鼠的全层皮肤切除动物模型,随机分为实验组和对照组,每组10只。实验组小鼠创面每天局部外用0.05g改为微孔多聚糖微粒,对照组不采取治疗。每天观察并记录两组创面大小及小鼠活动情况,比较创面愈合时间。术后12d处死所有小鼠,对创面肉芽组织进行组织学染色,量化并比较两组染色结果。结果实验组小鼠的创面愈合时间(8.9±0.3)d较对照组(10±0.4)d缩短,创面新生上皮(120.1±11.5)μm较对照组(96.6±5.9)μm增厚,胶原沉积率(0.55±0.01)%较对照组(0.48±0.02)%增加,ki-67阳性细胞(49±2)较对照组(44±2)增多、CD31阳性血管(17±1)较对照组(13±1)增多,经t检验差异均有统计学意义(P<0.05)。结论创面局部应用微孔多聚糖微粒可以缩短创面愈合时间,增加创面肉芽中胶原纤维的沉积,促进新生血管的增生,对创面局部的愈合具有促进作用。Obiective To investigate the effect on wound healing with the topical administration of MPH particle and do the preliminary discussion about the probable mechanism.Methods The full-thickness animal models of C57BL/6 were established. 20 mice were divided into two groups randomly, which are study group and control group. For the study group, 0.05 g MPH particle was administered to the wound area every day and no treatment was given to the control group. The wound size was measured and the parameters about mice like respiratory rate, weight were recorded every day. The time of wound closure for two groups was compared. On 12 day of post-operation, all the mice were sacrificed. The granulation tissues were stained by the technique of histopathology and the qualitative results of staining were compared. Results For the study group, the mean time of wound closure was reduced(8.9±0.3 vs 10±0.4)d. Compared with the control group, the staining results showed the epithelial thickness(120.1±11.5)μm vs ( 96.6±5.9)μm,collagen deposit index(0.55±0.01)% vs (0.48±0.02)%, the amount of ki-67 positive cells (49±2) vs (44±2)and CD31 positive vessels (17±1) vs (13±1)were found increased in the study group (P0.05). Conclusion The topical administration of MPH particle may accelerate and enhance wound healing.
