摘 要:目的讨论β-淀粉样蛋白前体(β-APP)脑缺血后变化规律并观察瑞香素对其影响。方法 wistar大鼠45只平分为三组:缺血再灌注组(简称对照组)分离双侧颈总动脉,无创微动脉夹夹闭双侧颈总动脉15min,然后恢复血流再灌注60min;再夹闭15min,松开动脉夹,恢复血流,缝合切口,术后每日用生理盐水灌胃;假手术组只分离双侧颈总动脉,不予夹闭,余同对照组;缺血再灌注瑞香素治疗组(简称治疗组)手术操作同对照组,每日用瑞香素灌胃。在2周,4周,6周不同的时间点取脑制作β-APP免疫组织化学切片,对β-APP表达阳性细胞计数。结果在2周,4周,6周不同的时间点,治疗组的阳性细胞数高于假手术组,低于对照组,不同时间点阳性细胞数呈递减趋势。假手术组的β-APP阳性细胞数无明显变化。对照组β-APP阳性细胞数最多。结论瑞香素对阻止β-APP的过度表达有一定的作用。Objective To analyze the changes of 15- amyloid protein precursor (β-APP) and how it was affected by daphnetin after cerebral isehemia. Methods Forty-five wistar rats were medially divided into three groups: a control group, a sham operation group and a therapy group. In the control group, the both side common carotid arteries were dissected and ligated for 15 minutes by artery clamp, were recoveried the blood supply for one hour, ligated again for 15 minutes, and then were reeoveried the blood supply by removing artery clamp. After operation the rats were lavaged by physiological saline daily. In the sham operation group, the both side common carotid arteries were only dissected without closure by clamp. Other processe was the same as control group. In therapy group, the operation was the same as control group, but the rats were lavaged by daphnetin differently. β-APP immunohistochemistry slices were made at the ends of second, 4th and 6th week and β-APP positive cells were counted. Results At the ends of second, 4th and 6th week, the positive β-APP cells in the therapy group were expressed more than that in the sham operation group, but less than that in the control group. The positive β-APP cells in the therapy group decreased gradually. There was no significant change in sham operation group. The control group has shown the most positive β-APP cells. Conclusions Daphnetin may play an important role in preventing cells from the excessive expression of β-APP.
