摘 要:通过灌胃硫酸铜溶液的方式模拟高铜摄入时铜对昆明小鼠动物血液生化指标的影响。50只30 d健康昆明小鼠随机分为5组,每组10只,雌雄各半。试验过程中,对照组、试验I、II、III组分别按硫酸铜0、20mg/kg、50mg/kg、100mg/kg体重灌胃,试验IV组按100mg/kg体重灌胃的同时饮用500 mg/L Vc溶液,其余各组自由饮用蒸馏水,试验持续4W。试验结束后,摘除眼球采血,常规分离血清后分别测定其AST、ALT、ALP等酶的活性和血清TP、Glu、TC、尿素、肌酐、钙、磷等含量。结果显示,与对照组相比,高铜灌胃小鼠可引起血清中AST、ALT、ALP、尿素、肌酐、钙离子等指标显著升高(P<0.05或P<0.01)。TP、Glu、TC等含量显著降低(P<0.05)。结果表明,高剂量铜能显著影响机体肝肾功能和机体的整体代谢状态。Vc能部分消除高铜摄入对机体的负面影响。This study was performed to explore the effect of high copper on serum biochemical indictors in Kun- ming mice with the graded level of copper sulfate solution by garage.. 50 30 - day - old Kunming mice were randomly divided into five groups ( n = 10) , and the ratio of male and female mice was 1 : 1. During the experiment, the control group, the test I, II, and III group were administrated with copper sulphate solution by gavage with graded levels of 0, 20 mg/kg, 50mg/kg, 100mg/kg body weight. All the mice except test group IV at the same dose as group III drinking distilled water containing 500mg/L Vc were supplied water ad libitum. The whole experimental period lasted for 4 weeks. At the end of the experiment, blood was collected by removing the eye balls. The collected blood was centrifuged for obtaining serum. The activities of AST, ALT, ALP, serum TP, Glu, TC, urea, creatinine, calcium, phosphorus and other routine indicators were determined. The results showed that, compared with the control group, the mice fed with high copper can cause serum activities of AST, ALT, ALP and urea, creatinine, calcium content increased significantly ( P 〈 0.05 or P 〈 0.01 ). TP, Glu, TC was significantly decreased (P 〈 0.05). These results showed that high doses of copper can significantly affect the body' s liver and kidney function and the body's overall metabolic state. Vc can partially eliminate the negative effects of high copper intake.
