摘  要:为研究促性腺激素抑制激素(Gonadotropin—inhibitory hormone,GnIH)基因表达与鹅卵泡发育间的关系,对鹅锄Ⅲ基因的CDS区序列进行了克隆与分析,并采用荧光定量PCR检测了该基因在产蛋高峰期鹅的不同大小卵泡中的表达量。序列分析结果表明,所获得的鹅翻删基因CDS区长471bp,编码157个氨基酸,与已知的其它鸟类GnlH相似性达87%。鹅锄坍前体包括3个“LPLRF”,具有典型的“LPXRF-酰胺”基序。荧光定量结果表明,鹅GnlH在所有不同大小的卵泡中均有表达,其表达量在卵泡发育过程中呈现规律性变化,总趋势是先增高后降低,且在直径4~5mm的卵泡中表达量最高。研究结果显示,鹅卵泡锄鹏基因的表达与卵泡选择性发育有关,且该选择过程可能发生在直径为4~5mm的卵泡。这一研究可为探讨季节性繁殖禽类卵泡发育的调控机理奠定基础。n order to discuss the relationship between gonadotropin-inhibitory hormone (GnIH)and development of follicles, the CDS of GnlH gene was amplified and analyzed. Its expression pattern was tested in different sizes of goose follicles at the peak of laying period by qRT-PCR. Sequence analysis results revealed that 471 bp of goose GnIH gene was obtained,which encoded 157 amino acids. The similarity between goose GnlH and those of other birds was 87%. In addition,goose GnlH precursor had three "LPLRF" ,which was the typical C-terminal "LPXRF-amide" of GnIH. Moreover, the result of qRT-PCR indicated that GnIH gene expressed in all the follicles detected, but its expression had a regular variation, which increased firstly,then decreased, and follicles with 4 to 5 mm in diameter had the highest expression level, revealing that the expression of goose GnIH gene had a relationship with selective development of follicles, and possibly it happened in those follicles with 4 to 5 mm in diameter. This research could make a foundation for discussing the mechanism of regulation about follicles development in seasonal breeding birds.
