摘 要:目的探讨脑梗死大鼠应用涤痰通腑汤对其脑缺血损伤的保护作用。方法 48只健康雄性SD大鼠随机分为假手术组、模型组、依达拉奉组和涤痰通腑汤组各12只。实验前10d各组大鼠进行高脂饮食制备痰热腑实证动物模型;实验第11天,除假手术组外,其余各组大鼠均制备局灶性脑缺血再灌注动物模型。观察术后6,24,48,72h各组大鼠神经功能行为学评分、测定术后72h大鼠血浆神经肽、脑组织肿瘤坏死因子-α和白细胞介素-1β水平。结果与假手术组比较,各模型组大鼠均出现明显神经功能缺损行为学表现,且大鼠脑组织肿瘤坏死因子-α、白细胞介素-1β和血浆神经肽水平均明显高于假手术组(P<0.05);手术后24,48h依达拉奉组和涤痰通腑汤组大鼠较模型组大鼠神经功能评分显著降低(P<0.05);手术后72h依达拉奉组和涤痰通腑汤组大鼠脑组织肿瘤坏死因子-α、白细胞介素-1β和血浆神经肽水平均较模型组大鼠降低(P<0.05)。结论涤痰通腑汤可能通过抑制炎性因子和血浆神经肽的产生而对大鼠脑缺血损伤起保护作用。Objective To study the protective effect of Ditan Tongfu decoction on rat cerebral infarction. Methods Forty-eight SD rats were randomly divided into sham operation group, model group, edaravone group and Ditan Tongfu decoction group, with 12 rats in each group. All rats were fed with high-fat diet for 10 days to prepare phlegm-heat accumulation models, and model group, edaravone group and Ditan Tongfu decoction group were established focal cerebral ischemia reperfusion models on the llth day. Neurological severity scores were observed in four groups 6, 24, 48 and 72 hours after operation. Plasma neuropeptide, tumor necrosis factovalpha (TNF-α ) and interleukin-1 beta (IL-1β) in brain tissue were detected 72 hours after operation. Results Compared with sham operation group, model group showed obvious neurological impairment. TNF-α and IL-1β in brain tissue and plasma neuropeptide were significantly higher in model group than those in sham operation group (P〈0.05). The neurological severity score was significantly lower in edaravone group and Ditan Tongfu decoction group than that in model group 24 and 48 hours after operation (P〈0.05). The levels of TNF-α, IL-1β in brain tissue and plasma neuropeptide were significantly lower in edaravone group and Ditan Tongfu decoction group than those in model group 72 hours after operation (P〈0.05). Conclusion Ditan Tongfu decoction plays an important role in preventing brain from ischemia damage by inhibiting production of inflammatory factor and plasma neuropeptide.
