摘  要:目的:对板栗花粉中的甾醇类物质进行提取、分离与成分鉴定,为花粉的开发奠定理论基础。方法:以乙醇为提取剂,提取温度70℃,提取时间为1.5h,固液比1:10对板栗花粉中的植物甾醇进行了提取,并用AB-8大孔树脂对其进行纯化,洗脱体系为80mL60%乙醇、60mL丙酮。使用N-甲基-N-(三甲基硅烷基)三氟乙酰胺活化物Ⅱ(MSTFAactivatedⅡ)为衍生化试剂,衍生后注入气相色谱-质谱联用仪器进行分析。研究结果显示得到了5种匹配度较高的甾醇,分别是:胆固醇、(3β,24R)-麦角甾-5-烯-3-醇、豆甾醇、β-谷甾醇和(3β)-豆甾-5,24(28)-双烯-3-醇。另使用溶剂提取法提取雌二醇,并用酶联免疫法(ELISA)对板栗花粉中雌二醇的含量进行了研究。结果显示雌二醇最大提取量为261.08pg/g。Phytosterols were extracted with ethanol from chestnut pollen(tempurate:70 ℃;time:1.5 h;solid-to-liquid ratio:1:10).The crude extract was purified by macroporous resin and analyzed by gas chromatograph-mass spectrometer(GC-MS) after the derivatization.N-methyl-N-(three methyl silicone alkyl) trifluoro ethyl amide activated Ⅱ(MSTFA activated Ⅱ) was used as the derivatization reagent.Different solvents were used to extract estradiol(E2).The result showed that from the purified phytosterols of chestnut pollen five kinds of sterols were isolated and identified,which were cholesterol,(3-beta,24R)-ergost-5-en-3-yl,Stigmasterol,beta-Sitosterol and(3beta)-stigmasta-5,24(28)-dien-3-yl.Estradiol was extracted by different solvents and detected by enzyme-linked immuno sorbent assay(ELISA).The result showed that maximum extraction of estradiol was 261.08 pg/g.
